Recommended Usage:

Each lot of this antibody is quality control tested by intracellular immunofluorescent staining using our nuclear factor staining protocol. For flow cytometric staining, the suggested use of this reagent is ≤0.125 µg per million cells in 100 µl volume. It is recommended that the reagent be titrated for optimal performance for each application.

* Alexa Fluor® 647 has a maximum emission of 668 nm when it is excited at 633 nm / 635 nm.

C57BL/6 mouse splenocytes were stained

C57BL/6 mouse splenocytes were stained with CD3 Alexa Fluor® 488, fixed and permeabilized with Nuclear Factor Fixation and Permeabilization Buffer Set , and then stained with Aiolos (clone 8B2) Alexa Fluor® 647 (top) or mouse IgG1, κ Alexa Fluor® 647 isotype control (bottom).

Description:

Aiolos, an Ikaros family member, is also known as Izkf3. It has a molecular weight of 58 kD and shows 70% homology to the Ikaros isoform, Ik-1, particularly in the DNA binding and C-terminal domains. Aiolos can form a heteromer with Ikaros or exist as a homodimer. The homomeric complex is a potent transcriptional activator.  Unlike Ikaros, Aiolos is expressed in late fetal thymus and adult lymphoid organs. Among the adult hematopoietic progenitors, Aiolos is expressed at low levels in mulitpotent but not in pluripotent stem cells. When these multipotent progenitors become more restricted to T and B lymphoid pathways there is an upregulation in Aiolos expression. The highest expression of Aiolos is detected in the spleen (especially in B-cells) followed by thymus and bone marrow, and cannot be detected in the non-lymphoid organs. Aiolos plays an important role in lymphocyte proliferation and differentiation.

Other Names: IKAROS family zinc finger 3 (Ikzf3), zinc finger protein Aiolos, Zfpn1a3, Znfn1a3
Structure: 509 amino acids with predicted MW of 58 kD
Distribution: Nucleus, cytoplasm
Restricted to lymphoid lineage, expressed in B, NK and T cells
Function: Transcription factor that plays an important role in the regulation of lymphocyte differentiation; plays an essential role in regulation of B-cell differentiation, proliferation, and maturation to an effector state
Interaction: Homodimer and heterodimer with other IKAROS family members; interacts with IKZF4, IKZF5, IKZF1, HRAS, FOXP3, and BCL21L isoform Bcl-X(L); associates with histone deacetylase complexes containing HDAC1, MTA2 and SIN3A
